Dysbiosis and inflammation in periodontitis: synergism and implications for treatment
نویسنده
چکیده
Recent advances from microbiome and host response mechanistic studies indicate that periodontitis is not a bacterial infection in the classical sense but rather results from dysbiosis owing to a breakdown in host-microbe homeostasis. Accordingly, periodontitis is induced in susceptible hosts by a polymicrobial community, in which different members have distinct roles that converge synergistically to enhance colonization, nutrient procurement and persistence in an inflammatory environment. Keystone pathogens – the colonization of which is facilitated by accessory pathogens – initially subvert the host response leading to the emergence of a dysbiotic microbiota, in which pathobionts overactivate the inflammatory response and cause periodontal tissue destruction. Inflammation and dysbiosis positively reinforce each other because inflammatory tissue breakdown products are used as nutrients by the dysbiotic microbiota, thereby contributing to its persistence and chronic inflammation. These concepts have important implications for novel therapeutic approaches, including strategies for microbial community manipulation and targeted modulation of the host response to limit destructive inflammation and reverse the microbial immune subversive tactics that fuel dysbiosis. Immunomicrobial pathogenesis of periodontitis: keystones, pathobionts, and host response.
Recent studies have uncovered novel mechanisms underlying the breakdown of periodontal host-microbe homeostasis, which can precipitate dysbiosis and periodontitis in susceptible hosts. Dysbiotic microbial communities of keystone pathogens and pathobionts are thought to exhibit synergistic virulence whereby not only can they endure the host response but can also thrive by exploiting tissue-destr...
متن کاملP94: Nigella Sativa Attenuates Neuro-Inflammation by Alleviating Periodontitis
Neuro-inflammation is a reaction to brain injury involving the activation of glial cells and release of inflammatory mediators, such as cytokines and chemokines which has a key role in neuro-degenerative diseases. Periodontitis is a microbiome-related dental inflammation caused by gram- negative bacteria that leads to connective tissue destruction and loss of teeth. Previous studies demonstrated...

Objectives: The purpose of this study was to evaluate the effect of low dose doxycycline and atorvastatin on gingival inflammation and alveolar bone loss in an experimental model of periodontitis in rats. Methods: Forty male Sprague Dawley rats were divided into four study groups as follows: (I) experimental periodontitis control, (II) rats with periodontitis treated with low dose atorvastatin...
متن کاملProspects for treatment of Porphyromonas gingivalis-mediated disease – immune-based therapy
Chronic periodontitis is an inflammatory disease of the supporting tissues of the teeth associated with a polymicrobial biofilm (subgingival plaque) accreted to the tooth which results in destruction of the tooth's supporting tissues. A characteristic feature of the disease-associated plaque is the emergence of proteolytic species. One of these species, Porphyromonas gingivalis has recently bee...

Neuro-inflammation, biochemical and cellular responses of the nervous system to injury, infection or neurodegenerative diseases, is a major risk factor for Alzheimer's, Parkinson's and Multiple Sclerosis. Periodontitis is a serious bacterial gum infection that damages the soft tissue and teeth supporting bones, causing teeth loss. It also renders systemic inflammation by secretion of pro-inflam...
